Recently the first total synthesis of the two natural depsipeptide Gq protein inhibitors YM-254890 and FR900359 was published by researchers at the University of Copenhagen, including the Molecular Pharmacology group. This new synthetic pathway has enabled us to develop structural analogues that could harbour activity at other G protein families such as Gs and G12/13. In my PhD project, I pharmacologically characterise novel analogues synthesised by our collaborators and investigate their binding using site-directed mutagenesis on the different G protein subtypes. The ultimate aim is to apply eventual inhibitors for the Gs and G12/13 proteins to delineate signalling pathways for less well-understood G protein coupled receptors.
