600 MHz Bruker Avance lll (NMR-600-b)

This is a 600 MHz Bruker ultra-shielded standard-bore magnet equipped with a 1.7-mm cryogenically cooled 1H/13C/15N TCI probe head with the highest possible mass sensitivity for 1H. The probe head is designed for 1.7-mm capillary sample tubes. This spectrometer is part of a state-of-the-art HPLC-PDA-HRMS-SPE-NMR instrument, that hyphenates analytical-scale HPLC-separation with photodiode array detection and high-resolution electrospray ionization mass spectrometry detection. Chromatographic traces (UV, TIC, BPC and/or EIC) are used for trapping of selected analytes using an automated solid-phase extraction unit after post-column dilution with water, and after repeated trapping of repeated separations, the trapped analyses are dried with pressurised nitrogen gas. The dried material is subsequently eluted with 30 µL of deuterated solvent to the volume-matched 1.7-mm NMR tubes in 96-tube rack systems using a Gilson autosampler with a 1-mm needle for tube filling. The HPLC-PDA-HRMS-SPE-NMR thus provides fully automated trapping and acquisition of UV, HRMS, and 1D and 2D NMR data directly from analytical-scale separation of crude extracts.
